I'm not sure if this is the same, but in MAME32, to configure the controls, just press 'Tab' while playing a game.

Defaults are something like:

 

1P Start = 1

1P Coin = 5

1P Button 1 = Control

1P Button 2 = Alt

1P Button 3 = Space

...

etc.

 

But yeah, check the TAB menu.

Because it's just like an arcade game, press 5 to insert a "coin", then 1 to start the game. Usually Left-Ctrl to fire.
